What prenatal formulas must exclude and balance
Prenatal formulation is an exclusion exercise
A prenatal is defined as much by what it leaves out as by what it contains. Upper limits are hard rather than advisory, several routine supplement ingredients are contraindicated in pregnancy, and two of the core nutrients interfere with each other. Most of the formulation work is exclusion and sequencing.
Preformed vitamin A
Retinol is teratogenic above roughly 3,000 mcg RAE a day, so prenatals carry beta-carotene instead. Any fish liver oil, liver extract or off-the-shelf multivitamin premix in the blend has to be checked for retinol carry-in.
Iron form and tolerance
Ferrous sulphate and fumarate deliver elemental iron cheaply but cause the constipation and nausea that make women stop taking a prenatal. Iron bisglycinate is gentler at the same elemental load and costs several times more.
Calcium blocks iron
Calcium at prenatal levels measurably reduces non-haem iron absorption when the two are taken together. Formulas that carry both either split them across a morning and an evening unit, or accept that the iron on the label is not the iron absorbed.
DHA source and oxidation
Prenatal DHA is usually 200 mg or more per day. Algal DHA avoids fish odour and cuts the heavy-metal testing burden. Fish oil is cheaper but needs a TOTOX limit and oxygen control during encapsulation.
The exclusion list
Ingredients that are routine elsewhere are barred here: high-dose retinol, most standardised herbal extracts, senna, high caffeine, and anything with traditional uterine-stimulant use. On a prenatal the exclusion list is usually longer than the active list.
Iodine and choline
Iodine is set around 150 to 220 mcg from potassium iodide. Choline is most often dropped: pregnancy adequate intake is 450 mg and bitartrate is about 40 percent choline, so a real dose is roughly a gram of powder, more than a tablet holds.
Formats
A prenatal rarely fits one dosage form. Iron and folate belong in a coated tablet, DHA needs a softgel, and putting an oil and a mineral in the same unit shortens shelf life. Two-part daily packs are the usual answer. Coating is not cosmetic here: uncoated iron tastes metallic and darkens on the shelf.
Halal & sourcing
The softgel is where a prenatal usually fails halal review, because DHA needs one and most shells are bovine or porcine gelatin. Fish gelatin or a starch shell solves it. Vitamin D3 is normally lanolin-derived, which is worth confirming rather than assuming.
