Formulating berberine: dose, form and tolerance
Berberine is a quaternary alkaloid extracted from Coptis chinensis, Phellodendron amurense or Berberis aristata. Almost every commercial product uses berberine HCl standardised to 97 to 98 per cent by HPLC. The formulation problem is not sourcing it. It is that berberine is bulky, bitter, bright yellow and barely absorbed, and those four properties decide the dose, the format and the shell.
Source and purity
Commercial berberine HCl trades at 97 to 98 per cent purity by HPLC. Ask for the botanical source on the certificate of analysis, because Coptis, Phellodendron and Berberis extracts carry different residual alkaloid profiles and different prices.
Split dosing
Commercial products run 500 mg per serving, taken two or three times daily. A single 1,500 mg dose is poorly tolerated and causes cramping and loose stools, so the pack has to support multi-dose use rather than one capsule a day.
Bioavailability strategies
Oral absorption sits under one per cent, because berberine is pumped straight back out by P-glycoprotein and cleared quickly in first pass. Formulators answer this with phytosome complexes, dihydroberberine at roughly a fifth of the dose, or co-dosed absorption aids.
Powder handling
Berberine HCl flows badly and compresses badly. A 500 mg fill needs a size 0 capsule and a glidant. A tablet usually needs granulation first, which adds a process step and a separate cost line to the quote.
Colour migration
Berberine stains. The yellow migrates into gelatin and HPMC shells and into white tablet coatings over shelf life, so clear capsules and pale coatings are a returns problem rather than a design decision.
Label cautions
Berberine inhibits CYP3A4, so interaction wording belongs on the carton. It is also standard practice to exclude pregnancy, breastfeeding and infants, since berberine displaces bilirubin and is contraindicated in jaundiced newborns.
Formats
Capsules are the default, because 500 mg of berberine HCl fills a size 0 cleanly and the shell hides the taste entirely. Tablets work if you accept the granulation step. Sachets rarely do, since the bitterness needs heavy masking. Capsules and tablets can start from as few as 100 boxes, so berberine is a cheap first SKU to test.
Halal & sourcing
Berberine itself is plant-derived, so the halal file turns on what surrounds it. Specify HPMC rather than bovine or porcine gelatin shells, and confirm the magnesium stearate comes from vegetable stearic acid rather than tallow.
